چکیده
Abstract R-loops are abundant and dynamic structures ubiquitously present in human cells both the nuclear mitochondrial genomes. They form cis wake of transcription complexes trans apart from complexes. In this review, we focus on relationship between topoisomerases, cancer genomics therapies. We summarize topological parameters associated with formation resolution R-loops, which absorb release high levels genomic negative supercoiling (Sc-). review deleterious consequences excessive rationalize how type IA (TOP3B) IB (TOP1) topoisomerases regulate resolve coordination helicase RNase H enzymes. also drugs (topoisomerase inhibitors, splicing G4 stabilizing ligands) predisposing genes (BRCA1/2, transcription, genes) known to induce whether thereby inducing damage can be viewed as a strategy for treatment.

Structural studies of type I topoisomerases
Topoisomerases are ubiquitous proteins found in all three domains of life. They change the topology of DNA via transient breaks on either one or two of the DNA strands to allow passage of another single or double DNA strand through the break. Topoisomerases are classified into two types: type I enzymes cleave one DNA strand and pass either one or two DNA strands through the break before reseali...
متن کاملATP inhibits nuclear and mitochondrial type I topoisomerases from human leukemia cells.
Type I topoisomerases have been purified from nuclei and mitochondria of human acute lymphoblastic leukemia cells. Both of these ATP-independent enzymes are actually found to be inhibited by ATP at physiologically significant concentrations. Other adenine nucleotides showed varying effects: ADP inhibited only at high concentrations; AMP had no effect on either topoisomerase. Both enzymes were a...
متن کاملR-loops in bacterial transcription
Nascent untranslated transcripts in bacteria are prone to generating RNA-DNA hybrids (R-loops); Rho-dependent transcription termination acts to reduce their prevalence. Here we discuss the mechanisms of R-loop formation and growth inhibition in bacteria.
متن کاملPhylogenomics of type II DNA topoisomerases.
Type II DNA topoisomerases (Topo II) are essential enzymes implicated in key nuclear processes. The recent discovery of a novel kind of Topo II (DNA topoisomerase VI) in Archaea led to a division of these enzymes into two non-homologous families, (Topo IIA and Topo IIB) and to the identification of the eukaryotic protein that initiates meiotic recombination, Spo11. In the present report, we hav...
متن کاملThe Effect of Dimethyl Sulfoxide on Supercoiled DNA Relaxation Catalyzed by Type I Topoisomerases
The effects of dimethyl sulfoxide (DMSO) on supercoiled plasmid DNA relaxation catalyzed by two typical type I topoisomerases were investigated in our studies. It is shown that DMSO in a low concentration (less than 20%, v/v) can induce a dose-related enhancement of the relaxation efficiency of Escherichia coli topoisomerase I (type IA). Conversely, obvious inhibitory effect on the activity of ...
